In 80 patients with hemoblastoses, expression of CD38, HLA-DR and CD143 (angiotensin-converting enzyme) was measured in PB and LC CD34+CD45low cells. The control group included 10 PB and 14 LC samples from healthy donors. Analysis of PB was performed prior to mobilization of hematopoietic stem cells (HSC) and on the day of leukapheresis prior to HSC collection. LC samples were examined at day 1 after HSC collection.<bold/></p><p><bold>Results.</bold> CD143 is expressed on CD34+CD45low cells both prior to mobilization and after it in all patients and donors, but CD34+CD45lowCD143+ cell counts varied depending on diagnosis and mobilization regimen. CD143+ expression on CD34+CD45low cells was significantly higher in patients who received combination of chemotherapy and granulocyte colony-stimulating factor compared to donors and patients with multiple myeloma who received only granulocyte colony-stimulating factor. Along with elevated CD34+CD45low cell count after hematopoiesis stimulation, CD34+CD45lowCD143+ cell counts also increased. It was shown that mobilized HSC almost completely lacks a fraction of early CD34+CD45low progenitor cells not expressing CD38, HLA-DR. Prior to hematopoiesis stimulation among CD34+CD45low cells, CD38+HLADR–cell fractions are prevalent, but after mobilization CD38–HLA-DR+ cell counts increased. No differences between CD34+CD45lowCD143+cell counts in patients with multiple myeloma depending on disease status, sex, age or number of chemotherapy courses prior to HSC mobilizationwere observed.</p><p><bold>Conclusion</bold>. Expression of angiotensin-converting enzyme on CD34+ cells in PB before and after HSC mobilization and in LC was observed. The cell counts varied depending on diagnosis and mobilization regimen.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Цель исследования</bold> – изучение особенностей субпопуляционного состава пула мобилизованных стволовых кроветворных клеток в периферической крови (ПК) и лейкоконцентратах (ЛК) у взрослых больных с онкогематологической патологией и доноров.<bold>Материалы и методы</bold>. Экспрессию CD38, HLA-DR и CD143 (ангиотензинпревращающий фермент) определяли в клетках CD34+CD45low ПК и ЛК у 80 больных гемобластозами. В контрольную группу включены 10 образцов ПК и 14 образцов ЛК здоровых доноров. Исследование ПК проводили до мобилизации стволовых кроветворных клеток (СКК) и в день лейкафереза до процедуры сбора СКК. Образцы ЛК исследовали в 1‑й день сбора СКК.<bold>Результаты</bold>. Показано, что CD143 экспрессируется на клетках CD34+CD45low как перед мобилизацией, так и после нее у всех пациентов и доноров, но количество клеток CD34+CD45lowCD143+ различалось в зависимости от диагноза и режимов мобилизации. Экспрессия CD143+ на клетках CD34+CD45low была статистически значимо больше у больных, которым применяли режимы, сочетающие химиотерапию и гранулоцитарный колониестимулирующий фактор, чем у доноров и больных множественной миеломой, у которых использовали гранулоцитарный колониестимулирующий фактор в монорежиме. Наряду с повышением содержания клеток CD34+CD45low после стимуляции кроветворения увеличивалось количество клеток CD34+CD45lowCD143+. Показано, что среди мобилизованных СКК практически отсутствует фракция ранних клеток-предшественниц CD34+CD45low, не экспрессирующая CD38, HLA-DR. Перед стимуляцией кроветворения среди клеток крови CD34+CD45low преобладают фракции клеток CD38+HLADR – , тогда как после мобилизации увеличилось содержание клеток CD38 – HLA-DR+. Не установлены различия между содержанием клеток CD34+CD45lowCD143+ у больных множественной миеломой в зависимости от статуса заболевания, пола, возраста и количества курсов химиотерапии, предшествующих мобилизации СКК.<bold>Заключение</bold>. Выявлена экспрессия ангиотензинпревращающего фермента на клетках CD34+ в ПК до и после мобилизации СКК и в ЛК.
